LOWICRYL RESIN Lowicryl resins are highly cross-linked acrylate-based embedding resins, designed for use over a wide range of embedding conditions. These resins provide low viscosity at low temperatures. The K4M kit is usable to -35°C and is a polar (hydrophilic) medium, while the HM20 kit can be used down to -70°C and is nonpolar (hydrophobic). Lowicryl's K11M and HM23 have properties similar to K4M and HM20, but can be used at temperatures 20 degrees lower than others: K11M at -60°C, HM23 at least -80°C. All of these resins are photopolymerised by long wavelength (360nm) ultraviolet light. Since the initiation of the polymerisation is largely independent of temperatures, blocks may be polymerised at the same temperatures used for infiltration. The resin may also be chemically polymerised at 60°C. LR GOLD RESIN The London Resin Company's acrylic "LR Gold" is the resin of choice when the antigenicity of the tissue is quite fragile. LR Gold is cured by the blue fraction of white light and this is possible at low temperatures. LR Gold is especially designed for low temperature (-25oC) embedding of fresh or lightly aldehyde-fixed tissue. Choice of initiator will determine polymerisation method - UV to intense white light. A PVP concentration above 20% may improve EM morphology but can also result in very high viscosities at low temperatures. LR Gold is non-toxic and has a low viscosity. LR WHITE RESIN A very low viscosity (8cps), non-toxic resin, suitable for both light and electron microscopy. LR White is a polar monomer polyhydroxylated achromatic acrylic resin. Curable by heat or light. Sections of polymerised LR White resin are hydrophilic. This characteristic allows immunocytochemistry reagents and stains to easily penetrate the sections. Etching of sections is not necessary. LR White sections show minimal non-specific staining. Its low viscosity makes it ideal for infiltrating decalcified bone and teeth as well as plant tissue. It is available in medium and hard grade. Distance, time and high temperatures during shipping from the manufacturer would age catalysed resin too much, we purchase and ship LR White uncatalysed, but the material is supplied with catalyst (benzoyl peroxide) and detailed instructions, but read in the linked User Notes how to catalyse without ruining the whole bottle. Catalysed LR White keeps refrigerated for over one year and embedded specimens can be cured using heat only. For cold curing with light, accelerator must be added. Newman, G.R. MAY-GRUENWALD STAIN This stain is sometimes favoured over Jenner's and it too is a combination of Eosin Y and Methylene Blue. Jenner's versions required aging, but this chemically polychromed Romanowsky type stain may be used immediately after preparation. The Biological Staining Commission does not certify May-Gruenwald.
